Effect of Scalp Acupuncture on the Expression of IL-6 and IL-10 in Hippocampal CA1 Region of Acute MCAO Rats

<p style="text-align: center;">ZHANG Tingzhuo, DU Xiaozheng, WANG Jinhai, TIAN Liang ,YUAN Bo, LI Xinglan, ZHANG Fenfan, QIAO Xiang, WANG Yixin</p><p style="text-align: center;">(1 School of Acu-moxibustion and Massage, Gansu University of Traditional Chinese Medicine, Lanzhou</p><p style="text-align: center;">730000; 2 Department of Traditional Chinese Medicine, the Second Hospital of Lanzhou University,</p><p style="text-align: center;">Lanzhou 730030)</p><p style="text-align: justify;"><strong>Abstract</strong>: <strong>Objective</strong>: To detect the expression of scalp-acupuncture on the expression of IL-6 and IL-10 of hippocampal&nbsp; CA1&nbsp; region&nbsp; in&nbsp; cerebral&nbsp; ischemia&nbsp; (CI)&nbsp; rats,&nbsp; so&nbsp; as&nbsp; to&nbsp; investigate&nbsp; its&nbsp; mechanism&nbsp; underlying regulation&nbsp; of Inflammation&nbsp; of ischemia stroke.&nbsp; <strong>Methods</strong>: 40&nbsp; SD rats were randomly allocated to normal group, model group,&nbsp; scalp-acupuncture group&nbsp; and&nbsp; drug&nbsp; group(n=10&nbsp; in&nbsp; each group).&nbsp; The model of&nbsp; focal&nbsp; CI&nbsp; was established&nbsp; by&nbsp; middle&nbsp; cerebral&nbsp; artery&nbsp; occlusion(MCAO).&nbsp; Scalp-acupuncture&nbsp; stimulation&nbsp; was&nbsp; applied&nbsp; to bilateral&nbsp; Dingnieqianxiexian&nbsp; (MS6) and Dingniehouxiexian&nbsp; (MS7)&nbsp; once daily for 7 days. Drug group&nbsp; were injection of pyrrolidine dithiocarbamate 100mg.kg-1/d, once daily for 7 days. The expression of IL-6 and IL-10 was detected by immunohistochemistry. <strong>Results</strong>: Compared with the normal group the expression of IL-6 and&nbsp; IL-10&nbsp; in the hippocampal CA1 region significantly&nbsp; increased&nbsp; in the model group (P&lt;0.05),&nbsp; while the expression of IL-6 in the hippocampal CA1 region obviousiy decreased in both scalp-acupuncture and drug group compared with the model group but the expression of IL-10 increased(P&lt;0.05); Compared with drug group,&nbsp; the&nbsp; expression&nbsp; of&nbsp; IL-6&nbsp; and&nbsp; IL-10&nbsp; in&nbsp; hippocampal&nbsp; CA1&nbsp; neurons&nbsp; of&nbsp; rats&nbsp; was&nbsp; not&nbsp; statistically significant(P&gt;0.05).&nbsp; Conclusion:&nbsp; Scalp-acupuncture&nbsp; stimulation&nbsp; can&nbsp; increase&nbsp; the&nbsp; expression&nbsp; ofantiinflammatory factor IL-10,&nbsp; while&nbsp; decrease&nbsp; the expression of&nbsp; inflammatory factor IL-6&nbsp; of ischemia stroke,which may be one of the molecular mechanisms of acupoint stimulation of acute ischemic cerebrovascular disease.</p><p><strong>Key Words</strong>: Scalp-acupuncture, Ischemic stroke, IL-6; IL-10, Inflammatory response</p><p style="text-align: center;"><br/></p>
